bitter taste and taste receptors and your gut. So everyone
knows that you have taste on your tongue, and that
taste tells you whether food is good to eat or not.
So you put something sweet on your tongue, it's very nice.

You put something bitter on your tongue, you want to
split that out. Well, it turns out you have taste
receptors or taste buds all the way down your gut,

we were talking about amerasate. Can you tell the listeners
how amerasate was originally discovered in what prompted the search
for natural appetite regulating compounds.
Speaker 5 (13:14):
Yeah, so it all started in two thousand and nine
when the New Zealand government put out a call to
do basically looked for a plant based alternative to drugs
for weight loss and appetite management. Now, the reason why
the government did this was twofold. Firstly because we have,
similar to America, an obesity crisis with growing numbers of

overweight and obese people as well as a large amount
of adolescent obesity. And there was also no real good
pharmaceutical options at the time. So we'd had a few
different drugs available. So Supertua mean, which is almost like
a stimulant, which had recently been withdrawn from the market
in sort of two thousand and five due to potential
(13:58):
cardiac issues. And there was another drug that was available,
never available in Europe Soorian America, but it was available
in Europe which was a sort of inverse cannabinoid agonist,
and so it sort of worked like anti marijuana, but
it caused depressive thoughts. So the government wanted essentially something
natural and something that would be safer, so they put
(14:19):
out a call and Plant of Food Research, the government
organization that I work for, decided that it would be
a good idea essentially to target the gaser intestinal tract
because there's a series of hormones there that are very
effective at regulating appetite and they can do so with
only sort of minimal side effects. If you get some
side effects, that tend to only be gaser intestinal and
(14:40):
they don't occur that frequently. So we really put together
a research program based on that and we started looking
at a few different theories. The one which we really
had off was using bitterness to regulate appetite and targeting
that to the gut.

And Sarah, I want you to come in on this
because without the scientific, pacific research and bringing it to
market that component, how can you describe the development of
calicurb as a commercially related product.
Speaker 3 (15:17):
Sure, dr Adam I was presented this extract in two
thy seventeen and in my time within this industry. I
had never seen an extract with that amount of science
behind it which looked into This was long before izempic,
(15:40):
which looked which could address such a big category where
we could actually own it from source to shelf. So
I was absolutely entranced by the science and I was asked, well,
what would you.

Do with it?

And a bit like the man that bought the company,
I was like, I absolutely love it. So here and
started the journey of you know, I raised money, I
formed a team, and in two thousand and eight we
launched CALICU. But yeah, it was a science that absolutely
I'd never seen science like it.
Speaker 2 (16:19):
And doctor Walker, you touched briefly on the science behind it.
What is this clinical research saying in about the effectiveness
of amerisate.
Speaker 5 (16:30):
Yeah, yeah, So we did several scientifical research studies in
order to validate efficacy. Essentially, the first thing we did
was we really wanted to show that we had these
bitter taste systems and the gats are intestinal tracts, so
that the first st age of the research was to
go into hospital and to get people ask people to
(16:53):
volunteer bits of their intestinal tracts, so biopsies from allow
along their intestinal tract to identify where all of these
bitter tasting systems were in the gut and to figure
out how's best to target them. We had twenty eight
people take part of them. We got this very nice
map of the gastro intestinal track, so we knew exactly
(17:13):
where to target and really what to use. We then
did a series of in vitro screening essays so drug
discovery essentially all drug discovery type systems, where we looked
for what would be best at triggering these bitter taste
systems within the gastra intestinal tract.

And then once we found a.

Potent activator in this case, it was a hop extract
we called a maris et we ran a series of
acute clinical trials to really show that it could work
in people. That the first of these trials was showing
the mode of action. And what we were able to
show is if the maris ad extract was given an
hour before and eat till your full meal and then
(17:57):
a subsequent snack, that people voluntarily eat eighteen percent less food.
So it's a big decrease in voluntary food consumption. And
they did this because they were getting increased amounts of
gut hormones being released in response to the meal. And
these hormones included GLP one, which is what is all
of the ozempic and we'll go via based on, as
(18:21):
well as CCK and PYY. So those are essentially the
three major appetite suppressing hormones from the gut, and we
were able to stimulate them to a very high degree.
So we got a sixfold change in GLP one and
a sixfold change in CCK, and around a doubling of PYY.
Now these push the activity levels of these hormones into
(18:43):
the appetite suppression range. And more importantly than that, we
were able to mimic the natural cycles of these hormones.
So what was happening was you're getting a normal response
to a meal and it was going up and then
down again, so following this natural circadian rhythm. It was
just being boosted in extended a little bit, So unlike
a synthetic GLP one, which is stabilized and injected, we
(19:05):
were able to increase these levels with the very natural
profile and just extend how long people were feeling full full,
and also you know, decrease the hunger they had at
the time of the meal.
Speaker 2 (19:20):
Yeah, and so Calaicurb's key ingredient, a Mercede, is derived
from hops and it mimics GLP one. Sarah, can you
expand on doctor Walker how he was explaining how the
plant based approach differs from the synthetic GLP one drugs
like ozempic, especially in the terms of safety in long

term sustainability. How do we keep that weight loss off
and how does calicurb work in that regard.

Sure, so, kellacurb or the active ingredient, it super stimulates
the release of the appetite suppressing hormones in your gut.
So we're actually we're actually superstimulating your own GLP one,
CCK and p YY because we all release it. But

as Dr Walker said, this maraced is doing it at
six times the normal level. So this is what I'm doing.
So with a o zempac or what we call a
synthetic hormone, these are synthetic hormones that you inject in yourself.
Nothing wrong with them, but they are a synthetic hormone.

And when you inject in a synthetic hormone, of course
you're always going to decrease your natural hormone release because
a synthetic hormone is doing it for you and your
own natural hormones go, hey, you know what, I'm going
on holiday. We don't let them go on holiday. We
exercise them and we get them to release, and we

get them to do that when you need it, which
is if you take calicurb an hour or an hour
and a half before a main meal like lunch or
lunch or dinner, this will decrease your intake. So because it.

that Aha moment with Caliicurb. You know again, listeners, it
took fifteen years and thirty million in research to develop
and that pivotal discovery or that aha moment in the lab?

What what did you see that was going to revolutionize
weight management?
Speaker 5 (24:03):
Well, so I can probably cover this one. Yeah, So
there were a few of those moments, so we were
really fortunate and that we had, I guess three main
questions we were looking at, and that was did we
have the bitter taste receptors in the gut, could we
activate them in the lab, and could we see clinical effects?
And we had we had aha moments, really for all
(24:23):
of those. So the first thing was identifying those those
taste buds or taste receptors.

In the gut.
Speaker 5 (24:29):
No one had done, I guess in depth study really
looking at that when we started, so getting that those
results and seeing those receptors were there was a real
big thing for us. Secondly, when you look at the
cells that produced these glp ones, these natural hormones, they
don't like doing it in response to bitterness. So they're
(24:50):
basically the bodies designed to eat, and so to trigger
them to send that stop eating signal from the gut
to the brain is quite difficult.

And so really even getting the.

Hop extract to make the cells release these hormones in
the lab was a really well we can actually, we
can actually do this because virtually nothing we tried worked
and that's probably not surprising to people, because if you
put something slightly bitter, like a cup of coffee or something,
you don't suddenly feel full. We really discovered that the
bitterness had to be very, very potent to work, and

we could see that in the lab, and I guess
the final aha moment came when we had the clinical trials.
So we had the first clinical trial a bunch of
sort of young guys who were sitting there and they
were presented with a meal, and they were almost they
almost saw it as a change to see how much
they could eat, and then they'd start the meal and
(25:46):
then all of a sudden they just stop and they're like, oh,
I'm really full.
Speaker 6 (25:51):
It just suddenly I just don't want to eat anymore.
This is really weird.
Speaker 5 (25:54):
And so the participants were giving this feedback and almost
this surprise on their face that they just weren't able
to eat this food we'd given them. And so I
think it was then that we really knew that there
was there was and that was before we had all
the hormone results. Just that behavioral response from the participants
really showed us that we had something that could actually
work and could really help people.

Yeah.
Speaker 2 (26:16):
Yeah, And I've been taking Calicurb for a few months

We get hundreds of hundreds of feedback from from consumers
using it, but the biggest one we always back is,
which is almost as Ed said, the surprise. You take
the noise from my brain. You take the noise from
my head. So this constant food noise for people is

I want to eat, What am I going to eat?
Where am I going to eat?

And so on like that.

You know, yes, we do get weight loss, we get
a lot of feedback on that, but it's a food noise.
It's a quietening of that. And just my own AHA
moment would have been in twenty and seventeen, I had
some test capsules and I was out at dinner with
a group of women and because when in New Zealand,

we had a little leg of slow cooped lamb in
the middle of us and we kind of finished eating.
But normally I would sort of peck. You know, you'd finish,
you'd be talking, you'd pick at the crunchy bits. And
I'd had a calicub before, and I was just like,
I'm full, and I just couldn't eat anymore. And I
(28:03):
suppose that was like that, as Egg said, It's almost
this incredible surprise. I'm full and I don't want to
eat anymore. So yeah, I'm a huge fan of it.
But the feedback is fantastic. Read the food noise.
Speaker 2 (28:19):
I agree with that, doctor Walker. Some people they hear
natural appetites as present and they assume it's just another fat.
How do you respond to skeptics who question whether plant
based solutions can truly compete with pharmaceuticals.
Speaker 6 (28:38):
Well, I guess there's a couple of ways to look
at that.
Speaker 5 (28:40):
And firstly that probably most pharmaceuticals are in fact based
on plant compounds, at least initially, so around fifty percent
maybe more of them are. And when you look at
things like even met Foreman, which is a diabetes drug
that's based on a plant compound. You know, we have aspirin,
which is a based on plant compound as well, And

so there's a lot of evidence in the market and
through the scientific literature. The plant compounds are very effective,
so effective that pharmaceutical companies based their drug discovery on them.

I think the second point.

Is that, unlike I guess most nutraceuticals or supplements that
are on the market, there's a really large quantity of
scientific evidence to support this and it goes through multiple levels.
So we have not only indications from historical usage that
bitterness can be used to regulate appetites. We've got this
(29:37):
great historical usage in various cultures, so that in the
Kalahari and the Highlands of Scotland they'd use bitterness to
regulate appetite to suppress it. We also have a very
solid mode of action, so we've shown these hormones can
be released from the gut, and we've shown that they
can be released at levels that are known to regulate appetite.
So we're hitting sort of a biological threshold for regulation

of appetite. And we're also able to show really meaningful
behavioral changes across several studies, So we can show meaningful
decreases and appetite, and we can show meaning decreases and
cravings for food, and particularly noticeable cravings for sweet food.
Post a meal, which of course is an area when
people really struggle. So I think that the body of

scientific evidence, and not only a theory that makes sense
based on the historical data, but then a body of
scientific evidence, a layer upon layer of Yes, we know
this works. We know there are taste receptors in the gun,
we know they stimulate these hormones.

All right, Sarah, So, for listeners considering calicurb, what's your
number one tip to maximize the results, whether that's timing
or diet synergy. I know we've discussed before on the
show about intermit and fasting. Tell us what's your secret

to maximizing results.

Well, then, humor who wants to reduce their meal size
take calicub at least well an hour to an hour
and a half before lunch and before dinner. So what
you want to do is take it on an empty stomach,
and the reason for that is you want it to
pass the stomach and it releases in the upper digestive system.

So you do want it. If you take it on
a full stomach, it's not going to get down there.
So always on an empty stomach, and I usually have
a glass of water. Now, how are you going to
remind yourself of that? You know, it doesn't have to
be an absolute hour, So I kind of say, you know,
when you have that morning coffee, that mid morning coffee
about ten o'clock or ten thirty, just take it. Then

you know that three o'clock or four o'clock in the
afternoon when you're feeling like, oh, you know, I want
something to perk me up. You know, well, it's not
going to perk you up, but you will remember to
take your caloricub. Another good hack for that is, of course,
putting a reminder on your phone, so that's the easiest
way to take it. You and I have talked about

intermittent fasting. I intermittent fast mainly because I'm lazy. I
think it's just easy for me. But I take mine
in the morning at about eight or nine o'clock and
that'll see me right the way through to lunchtime. And
al eat good lunch, but I just won't eat too much.

And you said, set yourself a reminder on your phone,
and that's a that's a good clue. What I use
as a clue is I just listen to my stomach.
So I'm if I'm getting a little bit a little
bit hungry that mid afternoon, I go ahead and take
another capsule real easy. It sits on the stomach, you know,
just drink it with a little bit of water, and
I'm usually good. I eat a light dinner then, so
it helps with that not you know, you go all

day and you don't just go ahead and just eat
everything inside of that dinner meal. So that's that's how
I use it. And so, doctor Walker, where do you
